目的:观察低强度中频超声对人卵巢癌细胞的杀伤作用。方法:取对数生长期的HO8910细胞,分成实验组和假辐照组,以低强度中频超声(频率1.7MHz)辐照。用MTT法计算细胞抑制率;光镜观察细胞形态,Hoechst33258荧光染色法检测凋亡细胞。结果:低强度中频超声照射各组间细胞抑制率与对照组相比有统计学差异,光镜下可见细胞形态学改变,同时Hoechst33258荧光染色法观察到亮蓝凝聚的凋亡细胞。结论:低强度中频超声对人卵巢癌HO8910细胞体外生长有明显杀伤作用,并可诱导其凋亡。
Objective: To observe the killing effect of low intensity ultrasound on human ovarian cancer cells. Methods: HO8910 cells in logarithmic growth phase were divided into experimental group and sham irradiation group and irradiated with low intensity ultrasound (frequency 1.7MHz). Cell inhibition rate was calculated by MTT method. Cell morphology was observed by light microscope. Apoptotic cells were detected by Hoechst 33258 fluorescent staining. Results: The inhibitory rates of cells in low intensity IF irradiation group were statistically different from those in control group. The morphological changes of cells were observed under light microscope. At the same time, the apoptotic cells were observed by Hoechst33258 fluorescence staining. Conclusion: Low-intensity ultrasound can obviously kill HO8910 human ovarian cancer cells in vitro and induce apoptosis.
