目的对比格犬发情期与间情期ERβ基因在性腺组织器官的变化状况进行荧光定量,明确ERβ基因在发情期与间情期下丘脑-垂体-性腺轴上的表达量差异情况,为深入研究比格犬发情机制提供基础。方法作为调控生殖的关键基因,ERβ基因位于比格犬下丘脑-垂体-性腺轴上,因此分别取处于发情期和间情期的比格犬,提取下丘脑、垂体、卵巢和子宫的RNA,反转录后对ERβ基因mRNA的表达量进行荧光定量PCR检测。结果间情期比格犬卵巢、子宫、垂体、下丘脑内ERβ基因mRNA的表达量分别是发情期比格犬卵巢、子宫、垂体、下丘脑内ERβ基因mRNA表达量的0.35倍、0.17倍、0.44倍、0.43倍。结论 ERβ基因在处于发情期的比格犬下丘脑-垂体-性腺轴内表达量均上调。
OBJECTIVE: To quantify the ERβ gene expression in estrus and estrus of the beagle dogs during estrus and estrus, and determine the expression of ERβ in the hypothalamus-pituitary-gonad axis during estrus and estrus. Beagle estrus mechanism to provide the foundation. Methods As the key gene for reproductive regulation, ERβ gene was located on the hypothalamus-pituitary-gonadal axis of Beagle dogs. Therefore, Beagle dogs in estrus and estrus were extracted respectively to extract RNA of the hypothalamus, pituitary, ovary and uterus, After reverse transcription, the expression of ERβ mRNA was detected by real-time PCR. Results The expression of ERβ mRNA in ovary, uterus, pituitary and hypothalamus of beagle dogs during estrus period were 0.35 times and 0.17 times of that of ERβ gene in ovary, uterus, pituitary and hypothalamus of estrus, 0.44 times, 0.43 times. Conclusion ERβ gene is up-regulated in hypothalamus-pituitary-gonad axis of beagle dogs in estrus cycle.
