本文叙述了用反相色谱法,采用三元溶剂体系分离测定生物样品中三种腺苷酸.三元溶剂为水、甲醇、乙腈(80∶15∶5)含0.005MpicA.在室温下等度洗脱能使AMP,ADP,ATP标准物25min完全分离.大鼠肝组织提取液中的AMP,ADP,ATP也能与杂质完全分离.再现性好,并优于Harmson和Rao报道的方法.五次测定结果的变异系数为AMp1.9%,ADP4.5%,ATP6.5%.回收率为95%、101%、99%.该方法能对大鼠肝、心肌、骨胳肌等组织样品中腺苷酸同时进行准确的测定.
This paper describes the separation and determination of three kinds of adenosines in biological samples by reversed-phase chromatography with ternary solvent system.The ternary solvent contains 0.005MpicA in water, methanol and acetonitrile (80:15:5) AMP, ADP and ATP standards could be completely separated after 25 min of elution.The AMP, ADP and ATP in rat liver tissue were also completely separated from the impurities.The reproducibility was better than the method reported by Harmson and Rao. The coefficient of variation of the assay was AMp1.9%, ADP4.5% and ATP6.5%, and the recoveries were 95%, 101% and 99%, respectively.The method was able to detect tissue samples of liver, myocardium and skeletal muscle In the adenine simultaneously accurate determination.
