目的探讨清肺口服液含药血清对腺病毒3Ⅰ、7b感染的人胚肺成纤维细胞之转化生长因子β1(TGFβ1)、血小板衍生生长因子BB(PDGFBB)蛋白表达的影响。方法细胞分为正常细胞组、病毒对照组、空白血清组、利巴韦林组和含药血清组,除正常细胞组外,其余4组均以3Ⅰ、7b型腺病毒分别攻击之,并分别加入相应药物至各组细胞中;采用酶联免疫吸附(ELISA)法检测各组细胞上清液中TGFβ1、PDGFBB的含量。结果病毒对照组较正常细胞组细胞中TGFβ1、PDGFBB含量均明显增多(P<0.01),含药血清组比病毒对照组的细胞TGFβ1、PDGFBB含量显著降低(P<0.01)。结论(1)腺病毒感染可使人胚肺成纤维细胞TGFβ1、PDGFBB蛋白表达增高;(2)清肺口服液可降低TGFβ1、PDGFBB细胞因子的蛋白表达,这可能是其抗病毒作用的机制之一。
Objective To investigate the effect of Qingfei oral drug-containing serum on the expression of transforming growth factor-β1 (TGFβ1) and platelet-derived growth factor-BB (PDGFBB) protein in human embryonic lung fibroblasts infected with adenovirus 3I and 7b. Methods The cells were divided into normal cell group, virus control group, blank serum group, ribavirin group and drug-containing serum group. Except the normal cell group, the other 4 groups were challenged with 3I and 7b adenovirus respectively. The corresponding drugs were added to each group of cells; the contents of TGFβ1 and PDGFBB in the supernatant of each group were detected by enzyme-linked immunosorbent assay (ELISA). Results The levels of TGFβ1 and PDGFBB in the virus control group were significantly higher than those in the normal group (P<0.01). The contents of TGFβ1 and PDGFBB in the virus-containing control group were significantly lower than those in the virus control group (P<0.01). Conclusion (1) Adenovirus infection can increase the expression of TGFβ1 and PDGFBB protein in human embryonic lung fibroblasts; (2) Qingfei Oral Liquid can reduce the protein expression of TGFβ1 and PDGFBB cytokines, which may be the mechanism of antiviral effect. one.
