目的采用中红外光谱衰减全反射光谱技术(attenuated total reflectance,ATR)对玄参生品与炮制品中哈巴俄苷的含量进行快速无损的测定。方法以高效液相色谱为对照分析方法,测定玄参炮制前后哈巴俄苷的含量,运用偏最小二乘法(partialleast squares,PLS)建立ATR光谱与哈巴俄苷的HPLC分析值之间多元校正模型,并对炮制前后的未知样本进行预测。结果校正模型决定系数生品为r2=83.81,SEE和SEP分别为0.021 55和0.051 03;制品校正模型决定系数为r2=95.56,SEE和SEP分别为0.007 30和0.034 78。结论该法可以作为一种准确、快速、高效、无损的检测方法用于检测中药炮制前后有效成分的变化规律。
Objective To determine the content of harpagoside in Radix Scrophulariae and its processed products by rapid attenuated total reflectance spectroscopy (ATR). Methods HPLC was used as a control method to determine the content of Harpagoside before and after the processing of Radix Scrophulariaceae. A multivariate calibration model was established between the ATR spectra and HPLC analysis of the harpagoside by partial least squares (PLS) And predict the unknown samples before and after processing. The results of the calibration model determined that the coefficient of raw product was r2 = 83.81, SEE and SEP were 0.021 55 and 0.051 03 respectively. The coefficient of determination of product calibration model was r2 = 95.56, SEE and SEP were 0.007 30 and 0.034 78 respectively. Conclusion This method can be used as an accurate, rapid, efficient and nondestructive detection method for detecting the variation of active ingredients before and after processing of Chinese traditional medicine.
