目的研究正常睡眠和睡眠剥夺后大鼠下丘脑和大脑皮质食欲素原mRNA的表达。方法用转筒法剥夺大鼠的睡眠,用逆转录聚合酶链反应(RT PCR)检测正常睡眠和不同剥夺睡眠时间大鼠下丘脑和大脑皮质食欲素原mRNA的表达。结果大鼠下丘脑的食欲素原mRNA昼夜有明显变化(光暗期6 00的0 406±0 012和光亮期18 00的0 835±0 045相比,P<0 01)。而在大脑皮质没有明显的昼夜变化。正常大鼠下丘脑的食欲素原mRNA表达要高于大脑皮质;短时间的睡眠剥夺不影响下丘脑和大脑皮质食欲素原mRNA的表达(对照组0 406±0 012与剥夺4h0 398±0 034相比,P>0 05),而睡眠剥夺8h后下丘脑和大脑皮质食欲素原mRNA的表达明显增加(对照组0 406±0 012与剥夺8h0 518±0 043、剥夺12h0 636±0 047和剥夺24h0 827±0 042,P<0 01)。结论食欲素与睡眠之间有密切的关系,正常睡眠的控制有赖于下丘脑的食欲素的调节。
Objective To study the expression of orexin mRNA in hypothalamus and cerebral cortex of rats after normal sleep and sleep deprivation. Methods The rats were deprived of sleep by the tumbling method. The expression of orexin mRNA in the hypothalamus and cortex was detected by reverse transcription-polymerase chain reaction (RT-PCR) in normal sleep and sleep deprived rats. Results The orexin mRNA in the hypothalamus of rats changed significantly from day to night (0 406 ± 0 012 at 600 in the light dark period and 0 835 ± 0 045 at the bright 18 00, P <0.01). There was no significant diurnal change in the cerebral cortex. Normal rat hypothalamus orexin mRNA expression was higher than the cerebral cortex; short-term sleep deprivation does not affect the hypothalamus and cerebral cortex orexin mRNA expression (control group 0 406 ± 0 012 and deprivation 4h0 398 ± 0 034 (P <0.05). After 8h sleep deprivation, the expression of orexin mRNA in hypothalamus and cerebral cortex increased significantly (0 406 ± 0 012 in control group and 8 518 ± 0 043 in deprivation and 12h0 636 ± 0 047 in deprivation Deprivation 24h0 827 ± 0 042, P <0 01). Conclusion There is a close relationship between orexin and sleep. The control of normal sleep depends on the regulation of orexin in the hypothalamus.
