目的建立LC-MS联用测定血浆五味子乙素的方法,观察五味子乙素在Wistar大鼠体内的药代动力学作用。方法提取五味子果实中的有效成分,给予大鼠一次性灌胃,其后在不同时间收集大鼠血液样品,通过LC-MS方法测定大鼠血浆中五味子乙素的浓度,分析大鼠血浆中五味子乙素的浓度-时间数据,应用3p97药代动力学分析软件计算大鼠体内五味子乙素的药代动力学参数。结果本法简单快速,可应用于不同样品中五味子乙素含量的测定。五味子乙素的保留时间为2.96min,血浆平均最大浓度为1.01μg/mL,灌胃后达峰时间为5.65hr,浓度时间曲线下的平均面积为15.98mg/h.L,平均清除率为0.94μg/h。结论本方法可应用于不同样本中五味子乙素的检测和实验动物体内五味子乙素的药代动力学分析。
Objective To establish a method for the determination of schisandrin B in plasma by LC-MS and observe the pharmacokinetics of Schisandrin B in Wistar rats. Methods The active ingredients in Schisandra chinensis fruit were extracted, and rats were given intragastric administration once a day. Then blood samples of rats were collected at different times. The concentration of schisandrin B in rat plasma was determined by LC-MS method and the plasma Schisandra chinensis was analyzed. The concentration-time data of B were used to calculate the pharmacokinetic parameters of Schizandrin B in rats using 3p97 pharmacokinetic analysis software. Results The method is simple and rapid and can be applied to the determination of schisandrin B in different samples. The retention time of schisandrin B was 2.96 min, the average plasma maximum concentration was 1.01 μg/mL, the peak time after intragastric administration was 5.65 hr, the average area under the concentration-time curve was 15.98 mg/hL, and the average clearance rate was 0.94 μg/mL. h. Conclusion This method can be applied to the determination of schisandrin B in different samples and the pharmacokinetics analysis of schisandrin B in experimental animals.
