膜渗滤亲和层析法的建立及在纯化尿HCG中的初步应用

作者报道一种亲和层析法并将其用于纯化人尿绒毛膜促性腺激素(HCG)。该方法是采用硝酸纤维素膜(NCM)作固相支持物吸附的抗体,用负压使待提液渗滤 NCM,在滤过的同时待提液中的相应抗原不断结合于 NCM上预吸附的抗体,再将抗原从NCM上解离,从而得到高纯度的抗原。结果表明用以提纯的HCG纯度达PAGE电泳单条带,免疫活性为9000IU/mg。故认为该法简便有效,可用于亲和层析纯化蛋白质。 The authors report an affinity chromatography and use it to purify human chorionic gonadotropin (HCG). The method uses the nitrocellulose membrane (NCM) as the antibody adsorbed by the solid support, so as to diafilter the liquid to be leached with NCM by a negative pressure, and preferentially adsorb the corresponding antigens in the liquid to be leached simultaneously on the NCM while filtering, Of the antibody, and then the antigen dissociation from the NCM, resulting in high purity antigen.
