目的:研究扶正化瘀方对原代培养CCl_4纤维肝细胞胶原生成率及白蛋白(A1b)分泌功能的影响。方法:制备CCl_4肝纤维化模型,胶原酶原位灌流法分离纤维肝细胞,以~3H-TdR掺入、~3H-Proline掺入胶原酶消化、ELISA等方法研究扶正化瘀方的血清药理学作用。结果:纤维肝细胞其增殖功能、A1b分泌功能均较正常肝细胞低下,而胶原生成率则显著上升;扶正化瘀方药物血清能明显抑制纤维肝细胞增殖及其胶原生成率,促进纤维肝细胞A1b分泌量。结论:上述结果可能是该方抗肝纤维化的作用机制之一,并在一定程度上体现了该方扶正(促进白蛋白合成)与化瘀(抑制胶原生成)的双重作用。
Objective: To study the effect of Fuzheng Huayu Recipe on collagen production rate and albumin (A1b) secretory function in primary cultured CCl_4 fibroblasts. METHODS: CCl_4 hepatic fibrosis model was established, fibroblast hepatocytes were isolated by collagenase in situ perfusion method, and the serum pharmacology of Fuzheng Huayu Decoction was studied by ~3H-TdR incorporation, ~3H-Proline incorporation, collagenase digestion, and ELISA. effect. Results: The proliferative function and A1b secretion function of fibroblasts were lower than those of normal hepatocytes, while the collagen production rate was significantly increased. Fuzheng Huayufang drug serum can significantly inhibit the proliferation of fibroblasts and collagen production rate, and promote fiber hepatocytes A1b secretion. Conclusion: The above results may be one of the mechanisms of the anti-hepatic fibrosis, and to a certain extent, it reflects the dual role of Fuzheng (promoting albumin synthesis) and pharmacology (inhibiting collagen production).
