探讨红杉醇对2型糖尿病大鼠肝病NADPH氧化酶亚单位p22 phox和p47 phox表达的影响。采用高脂高糖饮食加腹腔注射小剂量链脲佐菌素(streptozotocin,STZ)诱导糖尿病大鼠肝病模型,红杉醇(50、25及12.5 mg.kg 1.d 1)治疗6周。于末次给药后测定血糖、丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、总抗氧化能力(T-AOC)、过氧化氢(H2O2)、NO和胰岛素(Ins)的含量;real-time PCR测定肝脏p22 phox和p47phox mRNA表达;Western blotting法测定p22 phox和p47 phox蛋白表达,HE染色观察肝脏病理变化。结果发现,红杉醇各剂量组能降低模型大鼠空腹血糖、ALT、AST、Ins和H2O2含量,恢复胰岛素敏感指数(ISI)及体重,升高肝组织T-AOC和NO含量;降低肝组织NADPH氧化酶亚单位p22 phox和p47 phox mRNA及蛋白表达,改善肝细胞病理改变(水肿、点状或灶状坏死及炎症细胞浸润)。上述结果表明,红杉醇可通过降低NADPH氧化酶表达进而改善2型糖尿病大鼠肝病的氧化应激损伤。
To investigate the effect of cetuximab on the expression of NADPH oxidase subunits p22 phox and p47 phox in type 2 diabetic rats with liver disease. High-fat and high-sugar diet plus low-dose streptozotocin (STZ) intraperitoneal injection was used to induce diabetic rat models of liver disease and treated with 50, 25 and 12.5 mg · kg -1d 1 for 6 weeks. Blood glucose, alanine aminotransferase (ALT), aspartate aminotransferase (AST), total antioxidant capacity (T-AOC), hydrogen peroxide (H2O2), NO and insulin ( Ins). The expression of p22 phox and p47phox mRNA in liver was detected by real-time PCR. The expressions of p22 phox and p47 phox protein were detected by Western blotting. The pathological changes of liver were observed by HE staining. The results showed that each dose of cetyltrine could decrease fasting blood glucose, ALT, AST, Ins and H 2 O 2, restore insulin sensitivity index (ISI) and body weight, raise T-AOC and NO in liver tissue, NADPH oxidase subunits p22 phox and p47 phox mRNA and protein expression, improve liver cell pathological changes (edema, punctate or focal necrosis and inflammatory cell infiltration). The above results show that, by reducing the expression of NADPH oxidase, cetuximab can improve oxidative stress injury in liver of type 2 diabetic rats.
