采用高效液相色谱法在Hypersil NH2(300 mm×4.6 mm,5μm)上测定了发酵液中的四甲基吡嗪,流动相为乙腈-水(90︰10,V/V),流速为1 m L/min,紫外检测波长295 nm,柱温35℃。10 min内可以完成色谱检测。该方法的检出限(S/N=3)为0.03μg/L,线性范围0.02~2 g/L;方法回收率为96.7%~98.2%;RSD为0.54%~1.72%。试验结果证明,该方法是测定发酵液中四甲基吡嗪的快速、有效的定量分析方法。
The tetramethylpyrazine in the fermentation broth was determined by HPLC on Hypersil NH2 (300 mm × 4.6 mm, 5 μm) with acetonitrile-water (90:10, V / V) at a flow rate of 1 m L / min, UV detection wavelength 295 nm, column temperature 35 ℃. Chromatography can be completed within 10 min. The detection limit (S / N = 3) of this method was 0.03 μg / L and the linear range was 0.02-2 g / L. The recoveries were 96.7% -98.2%. The RSD was 0.54% -1.22%. The test results show that this method is a rapid and effective quantitative assay for the determination of tetramethylpyrazine in fermentation broth.
