目的:建立超高效液相法同时测定制剂中葛根素和毛蕊异黄酮葡萄糖苷含量。方法:采用UPLC方法,Waters symmetry C_(18)色谱柱(4.6×250 mm,5.0 um)以乙腈为流动相A,以0.2%甲酸水溶液为流动相B采用梯度洗脱的方式,体积流速为1ml/min,柱温35℃,检测波长260 nm。结果:葛根素与毛蕊异黄酮葡萄糖苷性范围分别为5.22~31.32μg/ml,0.974~24.3μg/ml,平均加样回收率分别为96.7%(RSD3.9%)、99.5%(RSD2.44%)。结论:该方法简单快捷,可用于黄芪制剂中毛蕊异黄酮葡萄糖苷和葛根素的同时测定,为制剂质量标准的建立提供了参考。
Objective: To establish a method for the simultaneous determination of puerarin and calycosin glucoside in preparations by ultra performance liquid chromatography. Methods: The mobile phase A was acetonitrile (acetonitrile), mobile phase B (0.2% formic acid) was used as the mobile phase B, and the volume flow rate was 1 ml on a Waters Symmetry C 18 column (4.6 × 250 mm, 5.0 μm) / min, column temperature 35 ℃, detection wavelength 260 nm. Results: The ranges of glucosidase of puerarin and calycosin were 5.22 ~ 31.32μg / ml and 0.974 ~ 24.3μg / ml respectively. The average recoveries were 96.7% (RSD 3.9%), 99.5% (RSD 2.44% ). Conclusion: The method is simple and rapid and can be used for the simultaneous determination of calycosin and puerarin in Astragalus membranaceus preparations, which provides a reference for the establishment of the quality standard.
