目的探讨蜜环菌多糖含药血清对过氧化氢(H2O2)诱导大鼠肾上腺嗜铬细胞瘤细胞(PC12细胞)损伤保护的作用机制。方法在H2O2诱导PC12细胞损伤模型的基础上加入蜜环菌多糖含药血清,采用MTT比色法检测含药血清对H2O2抑制PC12细胞增殖的影响;用比色法检测含药血清对H2O2诱导PC12细胞损伤培养液中乳酸脱氢酶的活力、丙二醛含量及超氧化物歧化酶活力的影响。结果不同浓度的含药血清对H2O2损伤的PC12细胞的存活率有提高作用,含药血清浓度在10%时达最大值(P<0.05或P<0.01);10%浓度的含药血清可明显降低模型细胞培养液中LDH活力、MDA含量,提高SOD活力。结论蜜环菌多糖含药血清对H2O2损伤的PC12细胞有保护作用,机制可能与提高细胞抗氧化能力有关。
Objective To investigate the protective effect of polysaccharides from Armillaria repellent polysaccharides on injury induced by hydrogen peroxide (H2O2) in rat adrenal pheochromocytoma cells (PC12 cells). Methods H2O2-induced PC12 cell injury model based on the addition of honey Armillaria polysaccharides serum containing MTT colorimetric assay containing H2O2 inhibited the proliferation of PC12 cells; using colorimetric assay of serum containing H2O2-induced PC12 Effects of Lactate Dehydrogenase Activity, Malondialdehyde Content and Superoxide Dismutase Activity in Cell Injured Culture Medium. Results The drug-containing serum at different concentrations had an effect on the survival rate of PC12 cells injured by H2O2. The serum concentration of drug-containing serum reached the maximum at 10% (P <0.05 or P <0.01). The serum containing 10% Reduce LDH activity and MDA content in model cell culture medium and increase SOD activity. Conclusion Armillaria polysaccharide-containing serum has a protective effect on PC12 cells injured by H2O2, and its mechanism may be related to the enhancement of cell antioxidant capacity.
