目的 研究前列腺素E2 (PGE2 )参与关节软骨退变的进程机理 ,以及肝细胞生长因子(HGF)对膝关节软骨损伤修复的作用机理。方法 对符合临床诊断标准的 30个膝关节 ,分别给予膝关节腔注射肝细胞生长因子 ,每天 1次 ,每次 2ml,观察PGE2 、尿中Ⅰ型胶原 (crosslaps)、碱性磷酸酶 (AKP)以及临床症状的改善程度 ,并做了给药 6周后与给药前的对比研究。结果 给药 6周后crosslaps和PGE2 与给药前比较均明显降低 (P <0 0 1,P <0 0 0 1)。给药 6周后临床症状均有不同程度的改善。结论 PGE2 参与了软骨损伤的某些阶段 ,肝细胞生长因子对膝关节软骨的修复有促进作用。
Objective To investigate the mechanism of prostaglandin E2 (PGE2) involved in the degeneration of articular cartilage and the mechanism of hepatocyte growth factor (HGF) repair of knee cartilage injury. Methods Thirty knee joints were enrolled in this study. Hepatocyte growth factor (HGF) was injected intraperitoneally into the knee joint. The rats were given 2 ml twice a day for 2 weeks. The levels of PGE2, type I collagen in urine, alkaline phosphatase (AKP) As well as the degree of improvement of clinical symptoms, and made the comparison of 6 weeks after administration and pre-administration. Results After 6 weeks of administration, the crosslaps and PGE2 significantly decreased compared with those before administration (P <0.01, P <0.01). After 6 weeks of treatment, clinical symptoms were improved to some extent. Conclusions PGE2 is involved in some stages of cartilage injury, and hepatocyte growth factor can promote the repair of knee cartilage.
