本文用原型胰岛素和A14“冷”单碘原子碘化胰岛素作标准品,同时用两株不同的豚鼠抗胰岛素抗血清作放射免疫分析。其结果经Scatchard分析和比较标准曲线斜率的方法证明,胰岛素分子A链第14位酪氨酸的碘标记,干扰其中一株抗血清(D7)固有的抗原抗体结合,降低其免疫活性,而对另一株抗血清(GP12)没有影响。此现象的意义在于进行自身置换分析等特殊的放射免疫分析时,必须考虑到~(125)碘标记对免疫活性或生物活性的干扰问题。
In this paper, the prototype insulin and A14 “cold” single iodine atom iodinated insulin as a standard, using two different guinea pig anti-insulin antiserum for radioimmunoassay. The results of Scatchard analysis and comparison of the slope of the standard curve proves that the 14th tyrosine of the A chain of insulin molecule is labeled and interferes with the antigen-antibody binding of one of the antisera (D7) to decrease its immunocompetence. However, The other antiserum (GP12) had no effect. The significance of this phenomenon is that special radioimmunoassay, such as self-replacement analysis, must take into account the perturbation of ~ (125) iodine labeling on immunocompetence or biological activity.
