以Iloprost液、生理盐水为保存液,对4℃保存兔胎脑皮层组织进行对比研究,以了解4℃保存的细胞存活率及能量代谢情况。结果表明:①随保存时间延长,细胞存活率逐渐下降,Iloprost液保存的细胞存活率明显高于生理盐水;②三磷酸腺苷(ATP)、能量负荷(EC)随保存时间延长明显下降,在保存2h后接近耗竭,两组间差异不显著(P>0.05)。从以上结果,我们认为Iloprost液的保存效果优于生理盐水,但保存时间仍有限,应考虑在细胞保护剂的基础上加用新陈代谢抑制剂,从而降低代谢速度,改善4℃保存
Iloprost solution and saline solution were used as the preservative solution to compare the preservation of rabbit cerebral cortex at 4 ℃ to find out the survival rate and energy metabolism at 4 ℃. The results showed that: (1) With the prolongation of storage time, the cell survival rate decreased gradually, and the cell survival rate of Iloprost solution was significantly higher than that of saline; (2) ATP, energy load (EC) decreased significantly with storage time, Nearly exhausted, there was no significant difference between the two groups (P> 0.05). From the above results, we believe that the preservation effect of Iloprost solution is better than that of saline solution, but the storage time is still limited. Consideration should be given to the use of cytoprotective agents based on the addition of metabolic inhibitors, thus reducing the metabolic rate and improving the preservation at 4 ℃
