目的通过检测酪氨酸激酶抑制剂AG490对前列腺癌DU-145细胞增殖及凋亡的影响,以期为前列腺癌的治疗奠定基础。方法将对数生长期的前列腺癌DU-145细胞随机分为AG490组和对照组。AG490组采用不同浓度的AG490干预,对照组不干预。干预24、48h后采用MTT法检测细胞生长抑制率。干预48h后流式细胞仪检测细胞凋亡率;采用免疫组化法检测Bcl-2表达。结果 AG490组的细胞生长抑制率和细胞凋亡率均明显高于对照组,呈时间和剂量依赖性,P均<0.05。AG490组的细胞Bcl-2蛋白表达明显低于对照组,P<0.05。结论 AG490可抑制DU-145细胞增殖、诱导其凋亡及减少Bcl-2的表达。
Objective To detect the effect of tyrosine kinase inhibitor AG490 on the proliferation and apoptosis of prostate cancer DU-145 cells in order to lay the foundation for the treatment of prostate cancer. Methods The logarithmic growth phase of prostate cancer DU-145 cells were randomly divided into AG490 group and control group. AG490 group with different concentrations of AG490 intervention, the control group did not interfere. After 24 and 48 hours of intervention, the cell growth inhibition rate was detected by MTT assay. Flow cytometry was used to detect the apoptosis rate 48h after intervention. The expression of Bcl-2 was detected by immunohistochemistry. Results The cell growth inhibition rate and apoptosis rate in AG490 group were significantly higher than those in control group, both in a time and dose-dependent manner (all P <0.05). AG490 cells Bcl-2 protein expression was significantly lower than the control group, P <0.05. Conclusion AG490 can inhibit the proliferation of DU-145 cells, induce apoptosis and decrease the expression of Bcl-2.
