目的在细胞水平研究1-(2,6-二甲基苯氧基)-2-(3,4-二甲氧基苯乙胺基)丙烷盐酸盐(DDPH)在低氧内皮细胞条件培养基(HECCM)促肺动脉平滑肌细胞(PASMC)的增殖过程中对c-fos和c-myc表达的影响。方法采用MTT比色法作为HECCM促PASMC的增殖指标,采用核酸斑点杂交法检测c-fos与c-myc mRNA的相对表达量,同时观察DDPH对它们的影响。结果低氧可刺激内皮细胞产生某些因子,经旁分泌作用促进PASMC的c-fos和c-myc基因转录以及促使PASMC增殖。DDPH能显著抑制HECCM促PASMC的增殖作用以及下调c-fos和c-myc mRNA的表达量。结论 DDPH能显著抑制HECCM促PASMC的增殖作用,其作用机制可能是通过下调C-fos和C-myc mRNA的表达量来实现的。
OBJECTIVE: To study the cell-level culture of 1- (2,6-dimethylphenoxy) -2- (3,4-dimethoxyphenethyl) propane hydrochloride (DDPH) in hypoxic endothelial cells (HECCM) on the proliferation of pulmonary artery smooth muscle cells (PASMC) on the expression of c-fos and c-myc. Methods MTT colorimetric method was used as the proliferation index of PASMC stimulated by HECCM. The relative expression of c-fos and c-myc mRNA was detected by dot blot hybridization, and the effects of DDPH on them were also observed. Results Hypoxia stimulated the endothelial cells to produce certain factors, promoted the transcription of c-fos and c-myc genes and promoted the proliferation of PASMC through paracrine effect. DDPH can significantly inhibit the proliferation of HECCM-induced PASMC and down-regulate the expression of c-fos and c-myc mRNA. Conclusions DDPH can significantly inhibit the proliferation of HECCM induced by PASMC. Its mechanism may be through down-regulating the expression of C-fos and C-myc mRNA.
