目的:探讨γ-氨基丁酸能神经元条件性敲除PGC-1α基因对小鼠焦虑抑郁行为的影响.方法:经实验室繁殖及基因型鉴定获得足够数量的PGC-1α基因敲除小鼠(纯合子小鼠),通过垂直开场实验、高架十字迷宫、强迫游泳以及悬尾实验评价条件性敲除PGC-1α基因对小鼠焦虑抑郁行为的影响.结果:垂直型开场行为学结果表明,纯合子小鼠开场行为活动显著高于杂合子(P<0.05)与野生型(P<0.01),运动、活动能力更强;高架十字迷宫实验结果表明,纯合子小鼠进入开放臂频率百分比显著高于杂合子和野生型小鼠(P<0.05),表现出抗焦虑特性;悬尾和强迫游泳实验结果表明,纯合子小鼠静止不动时间显著低于野生型小鼠(P<0.05),表现出抗抑郁特性.结论:γ-氨基丁酸能神经元条件性敲除PGC-1α基因使小鼠表现出抗焦虑和抗抑郁行为.“,”Objective:To test the effects of conditioned knockout of peroxisome proliferator-activated receptor-γ coactivator-1α(PGC-1α) gene in the GABAergic neuron on anxiety and depression in mice.Methods:The mice with conditioned knockout of PGC-1α gene in GABAnergic neurons were evaluated on the vertical open field and the elevated plus maze,as well as the tail suspension and forced swimming tasks.Results:The vertical open field and the elevated plus maze experiments showed that the homozygous mice were significantly hyperactive and less anxiety than the wild type ones (P < 0.05);the tail suspension and forced swimming tests exhibited that the homozygous mice show significantly anti-depressive performance than the wild type ones(P < 0.05).Conclusion:PGC-1α gene knockout in GABAnergic neurons might induce less-anxiety and anti-depression behavior in mice.
