目的:通过对甲状腺乳头状癌、结节性甲状腺肿患者及正常人群血清E-cadherin浓度的测定,结合E-cadherin蛋白在乳头状癌、结节性甲状腺肿和癌旁正常组织中的检测,分析E-cadherin在甲状腺乳头状癌中的作用及意义。方法:采用ELISA法检测E-cadherin在86例甲状腺乳头状癌、16例结节性甲状腺肿患者和10例正常人血清中的含量,并分析其与甲状腺乳头状癌临床病理特征的关系;应用Western Blot法检测E-cadherin蛋白在甲状腺乳头状癌、结节性甲状腺肿和癌旁正常组织中的含量。结果:甲状腺乳头状癌患者血清E-cadherin的含量明显高于结节性甲状腺肿患者和正常人(P<0.05),结节性甲状腺肿和正常人血清中E-cadherin的含量无明显差异(P>0.05)。E-cadherin蛋白在乳头状癌组织中的含量明显低于结节性甲状腺肿、癌旁组织(P<0.05),结节性甲状腺肿、癌旁组织中含量无明显差异(P>0.05)。血清中E-cadherin的高水平表达与甲状腺乳头状癌的临床病理分期和淋巴结转移相关(P<0.05),而与患者的性别、年龄及肿瘤直径大小不相关(P>0.05)。结论:血清E-cadherin上调与甲状腺乳头状癌的临床病理分期和远处淋巴结转移相关。患者血清中E-cadherin含量的检测,在甲状腺乳头状癌诊疗及预后方面具有重要意义。
OBJECTIVE: To determine the serum concentration of E-cadherin in papillary thyroid carcinoma, nodular goiter and normal human. Combined with the detection of E-cadherin protein in papillary carcinoma, nodular goiter and adjacent normal tissue, E-cadherin in papillary thyroid carcinoma and its significance. Methods: The content of E-cadherin in 86 cases of thyroid papillary carcinoma, 16 cases of nodular goiter and 10 cases of normal human serum was detected by ELISA. The relationship between the expression of E-cadherin and clinicopathological features of papillary thyroid carcinoma was analyzed. Western Blot assay E-cadherin protein in papillary thyroid carcinoma, nodular goiter and adjacent normal tissue content. Results: The serum level of E-cadherin in patients with papillary thyroid carcinoma was significantly higher than that in patients with nodular goiter and normal controls (P <0.05). There was no significant difference in serum E-cadherin between patients with nodular goiter and normal controls P> 0.05). The content of E-cadherin protein in papillary carcinoma tissues was significantly lower than that in nodular goiter, paracancer tissues (P <0.05), nodular goiter and paracancerous tissues (P> 0.05). The high level expression of E-cadherin in serum correlated with the clinicopathologic stage and lymph node metastasis of papillary thyroid carcinoma (P <0.05), but not with the gender, age and size of tumor (P> 0.05). Conclusion: The up-regulation of serum E-cadherin is associated with clinicopathologic stage and distant lymph node metastasis of papillary thyroid carcinoma. The detection of E-cadherin in patients’ serum is of great significance in the diagnosis and prognosis of papillary thyroid carcinoma.
