目的探讨康莱特注射液对胰腺癌荷瘤小鼠瘤体生长和瘤体周围微小淋巴道形成的抑制作用。方法体外培养Panc-02胰腺癌细胞株,构建C57BL/6小鼠皮下胰腺癌模型,随机分为5个组,每组8只。分别给予康莱特注射液低、中、高剂量、CTX和生理盐水腹腔注射,与给药后14d处死小鼠。测各组肿瘤大小,再对标本切片,行淋巴管内皮细胞透明质酸受体1(LYVE-1)免疫组化染色,计数各组LYVE-1标记的微淋巴管密度(MLD)。结果各用药组和NS组相比瘤体大小差异明显,且差异有统计学意义(p<0.05)。各组瘤体周边部的MLD明显高于瘤体中心区;康莱特中、高剂量组和生理盐水组瘤体周围MLD差异明显,且均有统计学意义(p<0.05)。结论康莱特注射液可以有效地抑制瘤体生长和瘤体周围淋巴管的生成。
Objective To investigate the inhibitory effect of Kanglaite Injection on the growth of tumor-bearing mice and the formation of lymph nodes around the tumor in tumor-bearing mice. Methods Panc-02 pancreatic cancer cell lines were cultured in vitro to establish a C57BL / 6 mouse model of subcutaneous pancreatic cancer and were randomly divided into 5 groups of 8 rats. Kanglaite injection was given low, medium and high doses, CTX and saline intraperitoneal injection, and mice were sacrificed 14d after administration. The size of tumor in each group was measured. The specimens were lysed by immunohistochemistry for LYVE-1 and lymphatic vessel density (MLD) of LYVE-1 in each group was counted. Results Compared with the NS group, the difference of the tumor size was significant and the difference was statistically significant (p <0.05). The MLD in the periphery of each group was significantly higher than that in the center of the tumor. There was significant difference in the MLD between the medium and high dose Kanglaite groups and the saline group (p <0.05). Conclusion Kanglaite injection can effectively inhibit tumor growth and lymphangiogenesis around the tumor.
