目的研究香菊胶囊溶液对单纯疱疹病毒2型的抗病毒作用,为生殖器疱疹的治疗寻找新的治疗手段。方法以Vero细胞为宿主细胞,阿昔洛韦为阳性对照药物,通过观察细胞病变效应(CPE)、噻唑蓝(MTT)比色法检测细胞存活率和病毒抑制率来评价香菊胶囊溶液体外对单纯疱疹病毒2型(HSV-2)的抑制作用。结果香菊胶囊溶液对Vero细胞的半数毒性浓度(TC_(50))为12.5mg/m L,药物和HSV-2共同作用2h后、病毒感染2h后加入药物、预防给药途径三种条件下对HSV-2的半数有效浓度(IC_(50))分别为1.901mg/m L,1.871mg/m L和1.942mg/m L,治疗指数(TI)分别为6.58,6.68和6.44,其抗病毒效果与阿昔洛韦差异无统计学意义(P>0.05)。结论香菊胶囊溶液在体外对HSV-2直接灭活作用明显,且对HSV-2的吸附和穿入细胞均有抑制作用,具有很好的体外抗疱疹病毒作用。
Objective To study the antiviral effect of Xiangju capsule solution on herpes simplex virus type 2 and to find new treatments for the treatment of genital herpes. Methods Vero cells were used as host cells and acyclovir was used as a positive control drug. Cell viability and virus inhibition rate were evaluated by observing cytopathic effect (CPE) and MTT colorimetric assay Inhibition of herpes simplex virus type 2 (HSV-2). Results The median toxicity concentration (TC_ (50)) of Xiangju Capsule solution to Vero cells was 12.5mg / m L, and the drugs and HSV-2 combined for 2h. The drugs were added after 2h of infection, The half effective concentration (IC 50) of HSV-2 were 1.901 mg / m L, 1.871 mg / m L and 1.942 mg / m L, respectively. The therapeutic indices (TI) were 6.58, 6.68 and 6.44, There was no significant difference between acyclovir and acyclovir (P> 0.05). Conclusion Xiangju capsule solution has obvious HSV-2 inactivation in vitro and has an inhibitory effect on HSV-2 adsorption and penetration into cells, and has good in vitro anti-herpesvirus activity.
