目的建立同时测定常山胡柚(Citrus Changshan-huyou Y.B.Chang)果汁中柚皮芸香苷、柚皮苷和新橙皮苷含量的方法。方法采用HPLC法。色谱柱Diamnosil C18柱(200 mm×4.6 mm,5μm);检测波长283 nm;流动相为甲醇-水-冰醋酸(体积比为37∶63∶0.2);流速1.0 mL.min-1;柱温为室温。结果柚皮芸香苷、柚皮苷和新橙皮苷质量浓度分别在72~576 mg.L-1、113~904 mg.L-1和53~424 mg.L-1内与峰面积呈良好的线性关系,回归方程分别为A=6.790×103ρ+1.457×104,r=0.999 6(n=6);A=1.267×104ρ-2.366×104,r=0.999 9(n=6)和A=1.110×104ρ-1.472×105,r=0.999 9(n=6)。平均回收率分别为98.3%、98.0%和98.4%。测得样品中柚皮芸香苷、柚皮苷和新橙皮苷含量分别为157、289和172 mg.L-1。结论方法准确、可靠,可为常山胡柚的质量控制提供参考依据。
Objective To establish a method for the simultaneous determination of rutin, naringin and neohesperidin in the fruit juice of Citrus Changshan-huyou Y.B.Chang. Methods HPLC method. The column was Diamnosil C18 (200 mm × 4.6 mm, 5 μm). The detection wavelength was 283 nm. The mobile phase was methanol-water-glacial acetic acid (volume ratio 37:63:0.2) For room temperature. Results The mass concentration of rutin, naringin and neohesperidin in grapefruit skin was good in 72 ~ 576 mg.L-1, 113 ~ 904 mg.L-1 and 53 ~ 424 mg.L-1, respectively The regression equation was A = 6.790 × 103ρ + 1.457 × 104, r = 0.999 6 (n = 6), A = 1.267 × 104ρ-2.366 × 104, r = 0.999 9 (n = 6) and A = 1.110 × 104ρ-1.472 × 105, r = 0.999 9 (n = 6). The average recoveries were 98.3%, 98.0% and 98.4%, respectively. The measured contents of pomelo rutin, naringin and neohesperidin were 157, 289 and 172 mg.L-1, respectively. Conclusion The method is accurate and reliable, which can provide reference for the quality control of Changshan grapefruit.
