【目的】探讨杜仲对大鼠骨髓间充质干细胞(BMSCs)增殖的影响。【方法】分别取10日龄SD大鼠胫骨、股骨的骨髓,过滤离心后将所得BMSCs进行体外单层培养、扩增,采用DMEM培养基,37℃、体积分数5%CO2条件培养。在细胞达到80%~90%融合时,使用2.5 g/L胰酶消化传代。将传代细胞分别置于空白血清培养液(对照组)、杜仲水提取物含药血清及杜仲醇提取物含药血清培养液进行培养,观察细胞在3种条件下的形态、贴壁生长、增殖、集落等情况,采用四甲基偶氮唑盐(MTT)法绘制生长曲线,比较3种培养条件下达到100%融合的时间。【结果】杜仲水提取物组、杜仲醇提取物组对大鼠BMSCs增殖的影响显著高于空白血清对照组(P<0.05),且杜仲醇提取物组作用优于杜仲水提取物组(P<0.05)。【结论】杜仲对大鼠BMSCs增殖具有一定促进作用。
【Objective】 To investigate the effect of Eucommia ulmoides Oliver on proliferation of rat bone marrow mesenchymal stem cells (BMSCs). 【Method】 Bone marrow of tibia and femur of 10-day-old SD rats were respectively taken out. After filtration and centrifugation, the obtained BMSCs were cultured in vitro monolayer and expanded in DMEM medium and cultured at 37 ℃ with 5% CO2. Passages were digested with 2.5 g / L trypsin when the cells reached 80% -90% confluency. The passage cells were placed in blank serum culture medium (control group), Eucommia ulmoides water extract serum and eucommia alcohol extract containing serum culture medium was cultured to observe the cells under three conditions morphology, adherent growth and proliferation , Colony and so on. The growth curve was drawn by MTT method and the time to reach 100% confluence was compared among the three culture conditions. [Result] The effects of Eucommia ulmoides extract group and Eucommia ulmoides alcohol extract group on the proliferation of BMSCs in rats were significantly higher than those in blank serum control group (P <0.05), and the effect of Eucommia ulmoides alcohol extract group was better than that of Eucommia ulmoides water extract group <0.05). 【Conclusion】 Eucommia ulmoides BMSCs can promote the proliferation of BMSCs in rats.
