10种胆汁酸在Minibeck试剂中,40C保温后各有特征性紫外吸收出现。一羟胆烷酸最大吸收峰在310nm处,二羟胆烷酸除在310nm处有一强吸收峰外,在390nm处还有一弱吸收峰;三羟胆烷酸在390nm处有强吸收峰,310nm处有一弱吸收峰。同一种胆烷酸其游离型与结合型在310或390nm处的ε值相近。胆汁酸浓度在5~10mM以下时与吸光率呈直线关系,在低温避光条件下至少可稳定48小时。三羟胆烷酸溶液中含有一羟或二羟胆烷酸时,在Minibeck试剂中的紫外吸收图形特征不变,最大吸收峰值有叠加特点,各组份胆汁酸量能通过测定310nm及390nm吸光度计算求得,故可用作体液胆汁酸分析的基础。
Ten kinds of bile acids in Minibeck reagent, each characteristic UV absorption appeared after 40C incubation. The maximum absorption peak of monohydroxycholanoic acid is at 310 nm. Dihydroxycholanoic acid has a strong absorption peak at 390 nm in addition to a strong absorption peak at 310 nm; trihydroxycholanoic acid has a strong absorption peak at 390 nm, 310 nm. There is a weak absorption peak. The same cholanoic acid has a similar ε value at 310 or 390 nm for its free and binding forms. When the bile acid concentration is 5 to 10 mM or less, it is in a linear relationship with the absorbance and is stable for at least 48 hours under low temperature and dark conditions. When the monohydroxyl or dihydroxycholanoic acid is contained in the trihydroxycholanoic acid solution, the UV absorption pattern characteristic in the Minibeck reagent is not changed, and the maximum absorption peak is superimposed. The bile acid amount of each component can determine the absorbance at 310 nm and 390 nm. Calculated and calculated, it can be used as the basis for bile acid analysis of body fluids.
