目的探讨核因子-κB(NF-κB)和c-myc在尖锐湿疣皮损角质形成细胞中的表达。方法采用原位杂交方法检测尖锐湿疣50例,确定为HPV 6/11阳性;采用免疫组化En Vision二步法检测50例尖锐湿疣患者皮损和25例正常人皮肤(包皮)中NF-κB和c-myc的表达及分布。结果①与正常人皮肤对照组相比,尖锐湿疣皮损角质形成细胞中NF-κB和c-myc的表达均有不同程度增强(前者U= 6.286,P<0.01;后者U=4.356,P<0.01)。NF-κB表达主要分布于棘细胞层,阳性信号定位于胞浆。c- myc表达主要分布于表皮全层或棘细胞层和基底层,阳性信号定位于胞核。②尖锐湿疣皮损中,NF-κB与c-myc的表达呈正相关(r=0.89,P<0.01)。结论在HPV6/11感染时,角质形成细胞中转录因子NF-κB和c-myc的表达增高,可能均参与HPV感染细胞增殖的调控。
Objective To investigate the expression of nuclear factor-κB (NF-κB) and c-myc in keratinocytes of condyloma acuminatum. Methods Fifty cases of condyloma acuminatum were detected by in situ hybridization (HPV 6/11). The immunohistochemical En Vision two-step method was used to detect the expression of NF-κB in 50 patients with condyloma acuminatum and 25 normal human skin (foreskin) And c-myc expression and distribution. Results ① Compared with the normal human skin control group, the expression of NF-κB and c-myc in keratinocytes of condyloma acuminatum increased to some extent (U = 6.286, P <0.01; the latter U = 4.356, P <0.01). NF-κB expression mainly in the spine cell layer, the positive signal localized in the cytoplasm. The expression of c-myc mainly distributed in the full-thickness epidermis or spine and basal lamina, the positive signal located in the nucleus. ② In condyloma acuminatum lesions, the expression of NF-κB and c-myc were positively correlated (r = 0.89, P <0.01). Conclusion The expression of NF-κB and c-myc in keratinocytes is increased in HPV6 / 11 infection, which may be involved in the regulation of HPV infection.
