目的:检测孕中期和孕晚期外周血和胎盘晚期糖基化终末产物(AGEs)及其受体RAGE、转化生长因子-β1(TGF-β1)的表达,探讨各指标与妊娠期高血压疾病的关系。方法:2014年8月至2015年10月在南通大学附属医院产科门诊产前检查做唐氏筛查的孕妇血清样本900例,追踪孕妇的妊娠结局,获取妊娠期高血压9例,子痫前期(PE)组6例。检测孕妇孕中期和孕晚期血清和胎盘中AGEs、RAGE、TGF-β1表达。结果:孕中期,PE组血清AGEs、RAGE、TGF-β1水平比对照组明显升高(P<0.05),妊娠期高血压组TGF-β1水平也较对照组明显升高(P<0.05)。孕晚期,妊娠期高血压组和PE组AGEs、TGF-β1水平比对照组均明显增高,PE组TGF-β1表达水平比妊娠期高血压组显著升高(P<0.05)。PE组RAGE表达水平比对照组和妊娠期高血压组均显著升高(P<0.05)。PE胎盘AGEs、RAGE、TGF-β1表达阳性染色细胞比对照组和妊娠期高血压组染色明显增强,数量明显增多。结论:AGEs-RAGE系统的激活可能和PE的发生密切相关,TGF-β1参与了这个病理过程。
Objective: To detect the expression of peripheral blood and advanced glycation end products (AGEs) and their receptors RAGE and TGF-β1 in the second and third trimester of pregnancy, Relationship. Methods: From August 2014 to October 2015, 900 serum samples of pregnant women who were tested for Down’s screening in obstetric clinic of Nantong University Affiliated Hospital were traced. The pregnant women’s pregnancy outcomes were tracked. Nine cases of gestational hypertension were acquired, and preeclampsia (PE) group of 6 cases. The levels of AGEs, RAGE and TGF-β1 in serum and placenta of pregnant women in the second and third trimester of pregnancy were detected. Results: In the second trimester of pregnancy, serum levels of AGEs, RAGE and TGF-β1 in PE group were significantly higher than those in control group (P <0.05), and levels of TGF-β1 in hypertensive pregnancy group were significantly higher than those in control group (P <0.05). In the third trimester of pregnancy, the levels of AGEs and TGF-β1 in hypertensive pregnancy group and PE group were significantly higher than those in control group. The expression of TGF-β1 in PE group was significantly higher than that in hypertensive pregnancy group (P <0.05). The expression of RAGE in PE group was significantly higher than that in control group and gestational hypertension group (P <0.05). The staining of AGEs, RAGE and TGF-β1 positive staining cells in PE placenta was more obvious than that in control group and gestational hypertension group. Conclusion: The activation of AGEs-RAGE system may be closely related to the occurrence of PE. TGF-β1 is involved in this pathological process.
