目的:测定不同产地半夏的氨基酸含量。方法:采用AccQ-Tag法,以AccQ-FlourTM对半夏中17个氨基酸进行柱前衍生;采用Waters 1525 HPLC仪器、AccQ-TagTM柱,以AccQ.Tag浓液-水(1∶10)为流动相A,乙腈-水(60∶40)为流动相B进行梯度洗脱,检测波长248 nm,柱温37℃,进样量10μL。结果:17个氨基酸的线性回归方程r值均>0.9997,平均回收率(n=6)为98.7%,RSD为1.8%;不同产地的半夏均含有17个氨基酸,不同产地的半夏总氨基酸含量以湖北产最高(19.18%),显著高于河南、贵州两产地(P<0.05),但与四川产地比较差异无统计学意义(P>0.05)。结论:本法快速、简便,结果准确,不同产地半夏总氨基酸含量差异较大。
Aims: To determine the amino acid content of Pinellia ternata in different habitats. Methods: AccQ-Tag method was used to pre-column derivatization of 17 amino acids in Pinellia ternata with AccQ-FlourTM. Waters 1525 HPLC instrument and AccQ-TagTM columns were run with AccQ.Tag concentrate-water (1:10) Phase A, acetonitrile-water (60:40) as mobile phase B gradient elution, detection wavelength 248 nm, the column temperature 37 ℃, the injection volume 10μL. Results: The linear regression equation of 17 amino acids were all r> 0.9997, the average recovery (n = 6) was 98.7% and the RSD was 1.8%. Pinellia ternata contained 17 amino acids in different producing areas, The content was the highest in Hubei (19.18%), which was significantly higher than that in Henan and Guizhou (P <0.05), but not significantly different from that in Sichuan (P> 0.05). Conclusion: The method is rapid, simple and accurate. The content of total amino acids in Pinellia ternata from different areas is quite different.
