目的:建立一种共表达绿色荧光蛋白( GFP)、红色荧光蛋白( RFP)和荧光素酶三报告基因稳定标记的骨肉瘤细胞株,并利用该细胞株建立骨肉瘤原位移植裸鼠模型,用于监测体内肿瘤的生长以及药物的评价。方法用慢病毒作为载体,将同时携带荧光蛋白和荧光素酶的基因转到U2-OS骨肉瘤细胞中,筛选稳定细胞株,将细胞原位移植到裸鼠的胫骨,通过活体成像观察骨肉瘤在裸鼠体内的生长转移。结果荧光标签能稳定地在细胞中表达,通过活体成像可观察到骨肉瘤在裸鼠体内的生长转移。结论成功建立了一种实时监测骨肉瘤在小鼠体内生长转移的模型,为骨肉瘤的机制研究和体内药物评价提供了一种理想的平台。“,”Objective To establish an orthotopic osteosarcoma nude mice model that co-expresses green fluorescent protein( GFP) , red fluorescent protein ( RFP) and luciferase for the purpose of monitoring the growth of osteosarcoma and screening drug candidates against osteosarcoma .Methods Human osteosarcoma cells of U 2-OS were infected with lentivirus carrying reporter gene .The reporter gene expression was verified by fluorescent microscopy and bioluminescence imaging.The cells were transplanted into tibia of the nude mice and monitored by bioluminescence imaging .Results The reporter gene was stably expressed in U 2-OS cells.The growth and metastsis of osteosarcoma could be detected in nude mice.Conclusion The established orthotopic osteosarcoma nude mice model is an ideal model for investigating the mechanism of growth and metastasis of osteosarcoma and for screening drug candidates against osteosarcoma .
