探讨鳖甲多肽的纯化、鉴定及其体外抗肝纤维化活性。通过超滤、葡聚糖凝胶Sephadex G-15及高效液相色谱法对鳖甲水解产物进行分离,得到一个纯化的寡肽(CTEPH-1:Asn-Pro-Asn-Pro-Thr)。采用MTS法和酶联免疫吸附法测定纯化多肽对活化肝星状细胞的抑制作用,发现通过降低各型胶原蛋白和TIMP-1的含量,CTEPH-1有效的抑制了肝星状细胞的活化和增殖。因此,该寡肽CTEPH-1可以作为功能食品的添加剂或潜在的抗肝纤维化药物。
Purification, Identification and Anti-Hepatic Fibrosis Activity of Biejia Polypeptide. A purified oligopeptide (CTEPH-1: Asn-Pro-Asn-Pro-Thr) was obtained by ultrafiltration, Sephadex G-15 and high performance liquid chromatography. The inhibitory effect of purified polypeptide on activated hepatic stellate cells was determined by MTS assay and enzyme-linked immunosorbent assay (ELISA). It was found that CTEPH-1 effectively inhibited the activation of hepatic stellate cells by decreasing the contents of various types of collagen and TIMP-1 proliferation. Therefore, the oligopeptide CTEPH-1 can be used as an additive in functional foods or a potential anti-hepatic fibrosis drug.
