目的研究肺动脉高压大鼠(Pulmonary Artery Hypertension,PAH)肺动脉平滑肌Ryanodine受体(Ryanodine Receptor,RyR)的功能。方法放射性配基法测定空白对照组、PAH的配体与受体平衡解离常数(K d)、最大结合容量(Bmax)。结果野百合碱(Monocrotaline,MCT)所致模型组RyR Bmax为(197±21.4)pmol·mg-1p较对照组(168±13.7)pmol·mg-1p明显增加(P<0.01);K d为(0.276±0.026)nmol/L与对照组(0.282±0.031)nmol/L相比无明显改变(P>0.05)。结论 PAH大鼠肺动脉平滑肌RyR数量发生了上调;受体的亲和力无改变。
Objective To investigate the function of Ryanodine receptor (RyR) in pulmonary arterial hypertension rats (PAH). Methods The radioligand method was used to determine the equilibrium dissociation constants (K d) and maximum binding capacity (Bmax) of PAH ligands and receptors in the blank control group. Results RyR Bmax was significantly higher (197 ± 21.4) pmol · mg-1p than that of the control group (168 ± 13.7 pmol · mg-1p) (P <0.01) (0.276 ± 0.026) nmol / L compared with the control group (0.282 ± 0.031) nmol / L (P> 0.05). Conclusions The number of RyR in pulmonary arterial smooth muscle of PAH rats is up-regulated; the affinity of the receptor is unchanged.
