本研究探讨中药复方君子汤(FFJZ)对抑制白血病K562细胞生长增殖和诱导凋亡的生物学特点及其可能的作用机制。应用细胞形态学、台盼蓝拒染法观察细胞生长状态;四氮甲唑蓝(MTT)法检测细胞增殖情况;流式细胞术检测细胞的凋亡率。结果表明:一定浓度范围的FFJZ作用于K562细胞后,随FFJZ药物浓度的升高其对K562细胞生长的抑制率明显增加(p<0.01),其半数抑制浓度(IC50)约为5.6mg/ml。在FFJZ浓度为4mg/ml时,细胞凋亡以早期凋亡为主,而至8mg/ml组时晚期凋亡比例显著增多,与未加药组比较有显著差异(p<0.01)。结论:在体外FFJZ一定浓度范围内可以抑制白血病K562细胞的生长增殖,诱导其凋亡并呈浓度依赖性,其机制可能与诱导细胞凋亡有关。
This study was aimed to investigate the biological characteristics and its possible mechanism of FFJZ on inhibiting proliferation and inducing apoptosis of K562 leukemia cells. Cell morphology was observed by cell morphology and trypan blue exclusion staining. Cell proliferation was measured by MTT assay. Cell apoptosis was detected by flow cytometry. The results showed that FFJZ could inhibit the growth of K562 cells with a dose-dependent increase of FFJZ concentration (p <0.01), and its IC50 was about 5.6mg / ml . At the FFJZ concentration of 4mg / ml, apoptosis was predominated by early apoptosis, while the rate of late apoptosis was significantly increased up to 8mg / ml, which was significantly different from that of the untreated group (p <0.01). Conclusion: FFJZ can inhibit the growth and proliferation of leukemia K562 cells in a concentration-dependent manner in vitro and induce its apoptosis in a concentration-dependent manner. The mechanism may be related to the induction of apoptosis.
