目的 制备基因重组人睫状神经营养因子 (rhCNTF)单克隆抗体。方法利用rhCNTF免疫BALB/C小鼠 ,取其脾细胞与小鼠NS -1骨髓瘤细胞融合 ,制备 3株抗rhCNTF的单克隆抗体 ,分别命名为CA2 、CA6 和BB11。结果 3株抗体类型均为IgG2a,免疫印迹试验证明为特异性抗CNTF的单克隆抗体 ;抗体相加试验表明 3株单抗是针对CNTF两个不同的抗原决定簇。
Objective To prepare monoclonal antibodies against human rhCNTF (recombinant human ciliary neurotrophic factor). Methods BALB / C mice were immunized with rhCNTF and their spleen cells were fused with mouse NS-1 myeloma cells. Three monoclonal antibodies against rhCNTF were prepared and named CA2, CA6 and BB11 respectively. Results The results showed that all the three antibodies were IgG2a, and their immunoblotting proved to be specific anti-CNTF monoclonal antibody. Antibody addition test showed that the three McAbs were directed against two different epitopes of CNTF.
