目的 探讨血清药理实验中血清供体动物的选择问题。方法 用MTT法检测不同种属的动物血清和人体自愿者的血清对 3T3细胞增殖的影响。结果 不同种属动物血清对培养细胞的细胞毒作用 ,似与其种属亲源有关。与其亲源最接近的动物血清最适于其生长 ,血清添加量达10 0 % ,尚可维持细胞正常生长 ,而与其亲源较远的动物 ,只有在低浓度下可维持细胞正常生长 ,血清加热灭活后细胞毒作用可以有所降低。结论 在选择含药血清的供体动物时 ,应尽量选择与培养细胞亲缘关系较近者
Objective To investigate the selection of serum donor animals in serum pharmacological experiments. Methods MTT assay was used to detect the effects of serum from different species of animal and human volunteers on the proliferation of 3T3 cells. Results Different species of animal serum cytotoxic effect on cultured cells, and its species may be related to the pro-source. The animal serum closest to its progeny was the most suitable for its growth. The amount of serum added was 100%. The normal growth of the cells was maintained. However, the animals far away from their ancestors maintained their normal growth only at low concentrations. Serum After inactivation of heat cytotoxicity can be reduced. Conclusion When selecting donor animals containing serum, we should try our best to choose the close relatives of cultured cells
