目的:探讨Ⅴ型胶原在肝纤维化中的作用。方法:以免疫组化技术和图像分析法对大鼠实验性肝纤维化时肝内Ⅰ、Ⅲ和Ⅴ型胶原(ColⅠ、ColⅢ和ColⅤ)的动态变化进行定量分析。结果:发现ColⅤ的变化和分布与ColⅠ和ColⅢ不尽相同:早期增加快于ColⅠ和ColⅢ,常分布于变性坏死的肝细胞周围;中期与ColⅠ和ColⅢ的增加同步;后期则渐趋回落。结论:在整个病变过程中ColⅤ具有多种细胞来源,它在病灶部位的沉积可能对促使肝纤维化的发生、病变的活跃进展具有重要意义
Objective: To investigate the role of type Ⅴ collagen in liver fibrosis. Methods: The dynamic changes of intrahepatic type Ⅰ, Ⅲ and type Ⅴ collagen (ColⅠ, Col Ⅲ and ColⅤ) in rats with experimental liver fibrosis were quantitatively analyzed by immunohistochemistry and image analysis. Results: The changes and distribution of ColⅤ were found to be different from that of ColⅠand Col Ⅲ: the early increase was faster than that of ColⅠ and Col Ⅲ, often distributed around the degenerated and necrotic hepatocytes; the increase of ColⅠ and Col Ⅲ was synchronous with that of ColⅠ and the later was gradually decreased. CONCLUSIONS: ColⅤ has multiple cell sources throughout the pathological process. Its deposition at the site of lesion may be of great importance for the occurrence of liver fibrosis and the active progression of the lesion
