目的观察扶正消瘤颗粒对人表皮生长因子受体2(HER-2)阳性乳腺癌细胞SKBR-3增殖和凋亡的影响。方法 SKBR-3细胞以2×106/2 000μL的密度接种到6孔培养板中,共5组。24 h后分别加入1倍、2倍、3倍剂量的扶正消瘤颗粒含药血清,以赫赛汀为阳性对照,空白血清为空白对照。加药后72 h,分别用四甲基偶氮唑蓝显色(MTT)法及流式细胞仪检测SKBR-3细胞的增殖和凋亡。结果 2倍剂量的扶正消瘤颗粒和赫赛汀均可抑制SKBR-3细胞增殖;3种剂量的扶正消瘤颗粒均可促进SKBR-3细胞早期凋亡,2倍剂量组早期凋亡率及总凋亡率最高。结论扶正消瘤颗粒可明显促进乳腺癌SKBR-3细胞凋亡,2倍剂量的扶正消瘤颗粒能抑制乳腺癌SKBR-3细胞增殖。
Objective To observe the effect of Fuzheng Xiaoyu Granule on the proliferation and apoptosis of human epidermal growth factor receptor 2 (HER-2) positive breast cancer cell line SKBR-3. Methods SKBR-3 cells were seeded into 6-well plates at a density of 2 × 10 6/2 000 μL for 5 groups. 24 h after adding 1 times, 2 times, 3 times the dose of Fuzheng Xiaoyu particles containing serum Herceptin as a positive control, blank serum blank control. 72 h after dosing, the proliferation and apoptosis of SKBR-3 cells were detected by MTT method and flow cytometry respectively. Results Both Fuzhengxiaojie granule and Herceptin could inhibit the proliferation of SKBR-3 cells at 2 times dose. Fuzhengxiaojie granules at 3 doses could promote the early apoptosis of SKBR-3 cells, The total apoptosis rate is highest. Conclusion Fuzheng Xiaoluowu Granule can obviously promote the apoptosis of SKBR-3 breast cancer cells. The 2-fold dose of Fuzheng Xiaoluowu Granule can inhibit the proliferation of breast cancer SKBR-3 cells.
