目的探讨miR-181a在肾透明细胞癌中的表达及miR-181a对肾透明细胞癌细胞系786-O功能的影响。方法通过q RT-PCR的方法检测miR-181a在42对肾透明细胞癌肿瘤及瘤旁组织和细胞系786-O、769-P、A498、Caki-1中的表达水平;以miR-181a mimics、miR-181a inhibitor转染786-O细胞系,通过MTS实验观察其增殖能力的改变,并通过流式细胞仪检测细胞周期及细胞凋亡的变化。结果 miR-181a在肾透明细胞癌中的表达明显高于瘤旁组织,同时其在细胞系786-O、769-P、A498、Caki-1中的表达明显高于其在人肾小管上皮细胞HKC中的表达。在786-O细胞系中,转染miR-181a mimics可以明显促进其增殖,诱导G1/S期转换并抑制凋亡;而miR-181a inhibitor则抑制增殖,诱导G1期阻滞并促进凋亡。结论 miR-181a在肾透明细胞癌中高表达并参与肿瘤的发生和发展,提示miR-181a可能成为肾癌治疗的潜在靶点。
Objective To investigate the expression of miR-181a in renal clear cell carcinoma and the effect of miR-181a on the function of renal clear cell carcinoma cell line 786-O. Methods miR-181a was detected by q RT-PCR in 42 pairs of renal clear cell carcinomas and their adjacent tissues and cell lines 786-O, 769-P, A498 and Caki-1. MiR-181a mimics MiR-181a inhibitor was transfected into 786-O cell line, and the proliferation of 786-O cells was observed by MTS assay. The cell cycle and apoptosis were detected by flow cytometry. Results The expression of miR-181a in renal clear cell carcinoma was significantly higher than that in tumor-adjacent tissues, and its expression in 786-O, 769-P, A498 and Caki-1 was significantly higher than that in human renal tubular epithelial cells HKC expression. In 786-O cell line, miR-181a mimics transfected miR-181a could significantly promote its proliferation, induce G1 / S phase transition and inhibit apoptosis, while miR-181a inhibitor inhibits proliferation, induces G1 arrest and promotes apoptosis. Conclusion miR-181a is highly expressed in renal clear cell carcinoma and is involved in the carcinogenesis and progression of tumors, suggesting that miR-181a may be a potential therapeutic target for renal cell carcinoma.
