目的探讨乳腺癌易感基因(BRCA1)蛋白在乳腺浸润性导管癌组织中的表达以及与临床病理的关系,了解BRCA1蛋白的表达与患者生存时间之间的相关性。方法采用SP免疫组化法对89例乳腺浸润性导管癌患者的石蜡标本进行BRCA1蛋白的检测,结合患者的年龄、病理组织学分级、肿块大小、腋淋巴结的转移情况等因素与生存时间进行相关性分析。结果BRCA1蛋白在乳腺癌的阳性表达率为94.3%(84/89),BRCA1蛋白的表达均定位于肿瘤细胞核,BRCA1蛋白的表达与年龄呈正相关,与肿块大小呈负相关,BRCA1蛋白的表达在不同组织学分级中(Ⅰ~Ⅲ级)差异有统计学意义(P<0.05),随着组织学分级的增加,BRCA1蛋白阳性表达的程度降低,生存时间减短,BRCA1蛋白的表达在有、无腋淋巴结转移的病例中差异有统计学意义(P<0.05),随着腋淋巴结数目的增加,BRCA1蛋白阳性表达的程度降低,生存时间也逐渐缩短。结论BRCA1蛋白的表达与年龄、肿块大小、组织学分级、腋淋巴结转移相关,随着进一步研究,推测BRCA1蛋白的检测将作为估计乳腺癌生物学行为和预后的新指标。
Objective To investigate the expression of breast cancer susceptibility gene (BRCA1) in breast invasive ductal carcinoma and its relationship with clinicopathological features and to find out the correlation between the expression of BRCA1 protein and the survival time of patients. Methods The SP immunohistochemical method was used to detect the expression of BRCA1 protein in 89 cases of invasive ductal carcinoma of the breast. The correlation of BRCA1 protein with the survival time was analyzed according to the age, histological grade, size of the tumor and the metastasis of axillary lymph node. Sexual analysis. With the increase of axillary lymph node number, the expression of BRCA1 protein decreased and the survival time decreased gradually. Conclusion The expression of BRCA1 protein is related to age, mass size, histological grade and axillary lymph node metastasis. With further study, it is speculated that the detection of BRCA1 protein will be a new indicator to evaluate the biological behavior and prognosis of breast cancer.
