目的了解神经干细胞巢蛋白(nestin)在新生大鼠缺氧缺血性脑损伤(HIBD)后表达的动态变化。方法56只新生7日龄SD大鼠随机分为假手术组、缺氧缺血组。制备大鼠HIBD模型后分别于即刻、3、12、24h,3、7、14d将动物处死。采用SABC免疫组化方法检测nestin蛋白表达的动态变化。结果假手术组nestin蛋白在海马、室管膜下区弱表达,在皮质几乎不表达,缺氧缺血组nestin蛋白12h在皮质,24h在海马、室管膜下区表达增加,nestin阳性细胞数分别为13.3±1.8,37.9±5.7,35.5±7.2,与假手术组相比有统计学差异,P<0.05。皮质、海马、室管膜下nestin阳性细胞数均在7d达高峰,分别为88.5±3.5,100.7±8.1,96.8±5.8,与假手术组相比有统计学差异,P<0.05,在缺血14d降低至假手术组水平。结论新生大鼠缺氧缺血脑损伤后nestin蛋白表达增加,nestin的表达可能是神经干细胞增殖及胶质细胞反应共同作用的结果。
Objective To investigate the dynamic changes of neural stem cells nestin after hypoxic-ischemic brain damage (HIBD) in neonatal rats. Methods 56 neonatal 7-day-old SD rats were randomly divided into sham operation group, hypoxic-ischemic group. Animals were sacrificed at 3,12,24h, 3,7,14d after preparation of HIBD model. The dynamic changes of the expression of nestin protein were detected by SABC immunohistochemistry. Results Nestin protein was weakly expressed in the hippocampus and subependymal zone of the sham-operated group, almost not expressed in the cortex. The expression of nestin protein in the hypoxic-ischemic group was increased at 12 h in the cortex and 24 h in the hippocampus and subependymal zone. The number of nestin-positive cells Respectively, 13.3 ± 1.8,37.9 ± 5.7,35.5 ± 7.2, compared with the sham-operated group, there was a significant difference (P <0.05). The number of nestin positive cells in the cortex, hippocampus and ependymal reached the peak at 7d, which were 88.5 ± 3.5, 100.7 ± 8.1 and 96.8 ± 5.8, respectively, which were significantly different from those in the sham operation group (P <0.05) 14d reduced to sham operation group level. Conclusion The expression of nestin increases after hypoxic-ischemic brain damage in neonatal rats, and the expression of nestin may be the result of the synergistic effect of neural stem cells proliferation and glial cell response.
