目的:建立大鼠血浆中葡萄内酯的检测方法,研究葡萄内酯在大鼠体内的药代动力学特性。方法:采用反相高效液相色谱法测定不同时间大鼠血浆中葡萄内酯的浓度,采用DAS 2.1软件计算药代动力学参数。结果:大鼠静脉注射葡萄内酯后AUC与给药剂量呈正比,Cmax与给药剂量基本成线性关系,MRT不随剂量增加而延长,但半衰期随剂量发生变化。结论:葡萄内酯经大鼠尾静脉注射后的体内过程符合二室模型,体内过程不完全符合线性动力学过程。
OBJECTIVE: To establish a method for the determination of grape lactone in rat plasma and to study the pharmacokinetics of grape lactone in rats. METHODS: The concentration of glucurolactone in rat plasma was determined by reversed-phase high performance liquid chromatography. DAS 2.1 software was used to calculate pharmacokinetic parameters. RESULTS: After intravenous administration of grape lactone, the AUC was directly proportional to the dose. The Cmax was basically linear with the dose. The MRT did not prolong with increasing dose, but the half-life changed with the dose. Conclusion: The in vivo process of grape lactone after tail vein injection is in accordance with the two-compartment model, and the in vivo process does not fully comply with the linear kinetic process.
