目的研究锰对大鼠肾上腺嗜铬瘤细胞(PC12)活性氧(ROS)的生成和miR-133b表达的影响。方法调整PC12细胞密度约为1×106/ml,以0(对照)、300、600μmol/L MnCl2分别染毒PC12细胞72 h,采用DCFH-DA荧光探针检测ROS生成情况;以0(对照)、100、300μmol/L MnCl2分别染毒PC12细胞24 h,采用荧光定量RT-PCR检测miR-133b相对表达水平。结果与对照组比较,300、600μmol/L MnCl2染毒组PC12细胞ROS水平升高(P<0.05);300μmol/L MnCl2染毒组PC12细胞miR-133b表达水平上调,而100μmol/L MnCl2染毒组PC12细胞miR-133b表达水平未见改变(P>0.05)。结论 MnCl2可诱导PC12细胞活性氧生成增加及miR-133b的表达上调。
Objective To study the effect of manganese on the generation of reactive oxygen species (ROS) and the expression of miR-133b in rat adrenal pheochromocytoma cells (PC12). Methods PC12 cells were treated with 0 (control) and 300,600 μmol / L MnCl2 for 72 h respectively. The density of PC12 cells was adjusted to 1 × 106 / ml, and the production of ROS was detected by DCFH-DA fluorescent probe. , PC12 cells were exposed to 100 and 300μmol / L MnCl2 for 24 hours respectively. The relative expression level of miR-133b was detected by RT-PCR. Results Compared with the control group, the level of ROS in PC12 cells exposed to 300 and 600μmol / L MnCl2 was increased (P <0.05). The expression of miR-133b was up-regulated in PC12 cells exposed to 300μmol / L MnCl2 and 100μmol / L MnCl2 The expression of miR-133b in PC12 cells was not changed (P> 0.05). Conclusion MnCl2 can induce the increase of reactive oxygen species production and the up-regulation of miR-133b in PC12 cells.
