目的观察DN大鼠肾脏解耦联蛋白2(UCP2)的表达,并探讨促红细胞生成素(EPO)对其表达的影响。方法 STZ构建糖尿病大鼠模型,随机分为糖尿病(DM)组,促红细胞生成素干预(EPO)组,另设健康对照(NC)组。PAS和HE染色观察各组肾脏病理变化,免疫组织化学及Western blot观察肾组织UCP2蛋白表达水平,RT-PCR测定各组肾组织UCP2mRNA表达。结果 DM组4、8、12周肾脏中UCP2mRNA及蛋白的表达逐渐升高,EPO组肾脏UCP2mRNA及蛋白表达均较DM组减少(P<0.05)。结论糖尿病大鼠肾脏中UCP2表达升高,EPO可减少UCP2表达,对肾脏起保护作用。
Objective To observe the expression of uncoupling protein 2 (UCP2) in the kidney of DN rats and investigate the effect of erythropoietin (EPO) on its expression. Methods The diabetic rat model was established by STZ. The diabetic rats were randomly divided into DM group, EPO group and NC group. The pathological changes of kidney were observed by PAS and HE staining. The expression of UCP2 protein in renal tissues was detected by immunohistochemistry and Western blot. The expression of UCP2 mRNA in renal tissues was detected by RT-PCR. Results The expression of UCP2 mRNA and protein in the kidney of DM group increased gradually at 4, 8 and 12 weeks, and the expression of UCP2 mRNA and protein in EPO group was lower than that of DM group (P <0.05). Conclusion The expression of UCP2 is increased in the kidney of diabetic rats. EPO can reduce the expression of UCP2 and protect the kidneys.
