目的探讨解偶联蛋白2(UCP-2)基因多态性与超重肥胖、2型糖尿病(T2DM)的关系。方法选取甘肃东乡族人群正常对照(NC)114名及T2DM患者104例,采用聚合酶链反应和变性高效液相色谱技术筛查UCP-2基因A1a55Val位点的基因型并测序。结果 AA、AV、VV基因型频率和A、V等位基因频率NC组和T2DM组比较,差异无统计学意义;两组中VV型、AV型者TG水平均高于AA型者,NC组VV型者BMI明显高于AA型者,差异均有统计学意义(P均<0.05)。结论 UCP-2基因A1a55Val的多态性可能与甘肃东乡族人群的超重肥胖及血脂异常相关,而与T2DM、胰岛素抵抗关系不大。
Objective To investigate the relationship between UCP-2 gene polymorphism and overweight and obesity, type 2 diabetes mellitus (T2DM). Methods 114 patients with normal controls (NC) and 104 patients with T2DM were selected from Dongxiang ethnic group of Gansu province. The genotypes of UCP-2 gene A1a55Val were screened by polymerase chain reaction and denaturing high performance liquid chromatography. Results The frequencies of AA, AV, VV genotypes and frequencies of A and V alleles were not significantly different between NC group and T2DM group. TG levels of VV type and AV type group were higher than those of AA type and NC group VV type BMI was significantly higher than AA type, the difference was statistically significant (P all <0.05). Conclusion The polymorphism of A1a55Val in UCP-2 gene may be related to overweight and obesity and dyslipidemia in Dongxiang ethnic group in Gansu province, but not to T2DM and insulin resistance.
