目的 研究放射联合高功率密度毫米波对肿瘤细胞的协同抗瘤效应。方法 用γ-射线、高功率密度毫米波单独或联合照射人舌鳞癌Tea 8113细胞,观察肿瘤细胞克隆形成抑制率,并行透射电镜和扫描电镜观察其超微结构改变。结果 放射联合高功率密度组(R+HL)抑瘤率82.0%,明显高于单放射组(R)的46.7%和单毫米波组(HL)的45.2%。毫米波功率密度、照射时间、有无联合放射对抑瘤率有非常显著的影响(P<0.01),放射与毫米波照射次序对抑瘤率无显著影响(P>M0.05)。电镜观察发现照射后细胞表面绒毛粘连、融合、脱落、细胞膜微孔、线粒体变性、胞浆内大量空泡等改变。结论 放射联合高功率密度毫米波对人舌鳞癌细胞有明显的协同抗瘤效应,放射与毫米波照射次序对抑瘤率无明显影响。
Objective To study the synergistic anti-tumor effect of radiotherapy combined with high power density millimeter wave on tumor cells. Methods Human tongue squamous cell carcinoma cell line 8113 was irradiated with γ-ray and high power density millimeter wave separately. The inhibition rate of tumor cell clone formation was observed. The ultrastructural changes were observed by transmission electron microscopy and scanning electron microscopy. Results The radiotherapy combined with high power density group (R + HL) had a tumor inhibition rate of 82.0%, which was significantly higher than 46.7% in single radiation group (R) and 45.2% in single millimeter wave group (HL). The power density of millimeter wave, irradiation time and presence or absence of co-irradiation had a significant effect on tumor inhibition rate (P <0.01). The order of irradiation and millimeter wave irradiation had no significant effect on tumor inhibition rate (P> 0.05). Electron microscopy showed that cell surface villus adhesion, fusion, shedding, cell membrane pores, mitochondrial degeneration, a large number of vacuoles and other changes in the cytoplasm after irradiation. Conclusion Radiation combined with high power density millimeter wave has obvious synergistic anti-tumor effect on human tongue squamous carcinoma cells. Radiation and millimeter wave irradiation have no significant effect on tumor inhibition rate.
