采用白血病祖细胞集落形成(CFUAML) 培养法,观察不同浓度人参总皂甙(TSPG) 对HL60、Meg01 白血病细胞株和急性髓细胞白血病患者的骨髓细胞集落形成的影响,并以正常粒单祖细胞(CFUGM)作比较。结果:正常和白血病祖细胞均对低浓度(5~25ug/ml) 的TSPG有反应,集落数提高26.1~35.4% ;中等浓度(75ug/ml) 的TSPG仍然刺激正常CFUGM 集落增加,但抑制HL60、Meg01 和CFUAML集落生长,抑制率分别为50.8% 、25 .1 % 和19.0% ;较高浓度(100ug/ml) 的TSPG对白血病祖细胞的抑制作用更明显,HL60 不能形成集落,对Meg01 和CFUAML的抑制率分别为53.3% 和28 .9 % ,而正常CFUGM 仍生长良好。提示TSPG对白血病祖细胞的影响不同于正常祖细胞,不同的浓度显示为增殖或抑制的双向作用
Using leukemia progenitor cell colony formation (CFU-AML) culture method, the effects of different concentrations of total saponin (TSPG) on bone marrow cell colony formation in HL60, Meg01 leukemia cell lines and acute myeloid leukemia patients were observed. Normal granulocyte-progenitor cells (CFU-GM) were compared. Results: Both normal and leukemic progenitor cells responded to low concentrations (5-25ug/ml) of TSPG and the number of colonies increased by 26.1 to 35.4%; moderate concentrations (75ug/ml) of TSPG still stimulated normal CFU-GM The colony increased, but the inhibition of colony growth of HL60, Meg01 and CFUAML was inhibited by 50.8% and 25, respectively. 1% and 19.0%; higher concentrations (100ug/ml) of TSPG significantly inhibited the leukemic progenitor cells, HL-60 could not form colonies, and the inhibition rates of Meg-01 and CFU-AML were 53. 3% and 28. 9 %, while normal CFU GM still grows well. It is suggested that the effect of TSPG on leukemia progenitor cells is different from that of normal progenitor cells, and different concentrations are shown as two-way effects of proliferation or inhibition
