目的探讨雷公藤红素对人肥大细胞白血病的作用及机制。方法以人肥大细胞白血病细胞系HMC1细胞为靶细胞,观察雷公藤红素对HMC1细胞凋亡的诱导作用。结果①0.12~1.00μmol/L雷公藤红素可以引起HMC1细胞凋亡,表现为细胞呈凋亡形态学改变,DNA琼脂糖凝胶电泳出现梯形条带,流式细胞仪分析HMC1细胞在G1期前出现凋亡峰;②凋亡率随药物浓度的增加及作用时间的延长而递增;③促进G1期细胞进入S期,使S期细胞比例升高,随后凋亡率升高,S期细胞下降,两者呈负相关(P<0.01);④雷公藤红素可使HMC1细胞bax、cmyc表达增加,bcl2表达下降。结论雷公藤红素可有效地诱导HMC1细胞凋亡,凋亡主要发生在S期,凋亡的发生与其上调促进凋亡基因bax、cmyc和下调抑凋亡基因bcl2的表达有关。
Objective To investigate the effect and mechanism of tripterine on human mast cell leukemia. Methods Human mast cell leukemia cell line HMC1 cells were used as target cells to observe the induction of apoptosis of HMC1 cells by tripterine. Results 10.12~1.00μmol/L tripterine could induce apoptosis of HMC1 cells, which showed apoptotic morphological changes. DNA ladders appeared on agarose gel electrophoresis, and flow cytometry was used to analyze HMC. Conclusion Tripterine can effectively induce the apoptosis of HMC-1 cells. The apoptosis mainly occurs in S phase.
