目的 本研究的目的是探讨基质金属蛋白酶 2 (matrixmetalloproteinase 2 ,MMP 2 )及基质金属蛋白酶 9(matrixmetalloproteinase 9,MMP 9)的表达与侵袭性垂体腺瘤相关性研究。以寻找垂体腺瘤侵袭性可能的发病机理。方法 用免疫组化方法对 5 4例垂体瘤患者的组织标本进行研究。并对其中 16例患者采用逆转录 聚合酶链反应 (RT PCR)的方法对MMP 2及MMP 9mRNA的表达进行分析。结果 5 4例垂体瘤患者中 ,有 32例女性 ,2 2例男性。平均年龄为 4 9.9岁 (从 18~ 76岁 )。其中 ,12例为侵袭性垂体腺瘤 ,4 2例为非侵袭性垂体腺瘤。免疫组化显示 ,侵袭性垂体腺瘤的MMP 2及MMP 9的表达明显高于非侵袭性垂体腺瘤 (3.9± 0 .5 ;2 .3± 0 .2 ;P <0 .0 1)和 (4.1± 0 .4 ;2 .6± 0 .2 ;P <0 .0 1)。MMP 2及MMP 9的表达与MIB 1的表达无明显相关性 (r=$C 0 .0 5 ;P >0 .0 5 )。侵袭性垂体腺瘤MMP 2及MMP 9mRNA的表达 ,明显高于非侵袭性垂体腺瘤 [(5 9.7± 12 .5 ) % ;(33.3± 5 .4 ) % ,P <0 .0 5 ]及 [(6 8.2± 15 .3) % ;(2 1.8± 8.2 ) % ;P <0 .0 5 ]。结论 MMP 2及MMP 9的高表达与肿瘤的侵袭性密切相关。但与肿瘤的大小及分泌功能无明显关系。MMP 2及MMP 9可以作为肿瘤侵袭性一项有效的指标。肿瘤的侵袭性和增殖之
Objectives The purpose of this study was to investigate the relationship between matrix metalloproteinase 2 (MMP 2) and matrix metalloproteinase 9 (MMP 9) expression in invasive pituitary adenomas. In search of the possible pathogenesis of invasive pituitary adenomas. Methods Immunohistochemistry was used to study the tissue samples from 54 patients with pituitary adenoma. The expression of MMP-2 and MMP-9 mRNA in 16 of them was analyzed by reverse transcription-polymerase chain reaction (RT-PCR). Results Among 54 cases of pituitary adenomas, 32 were female and 22 were male. The average age was 49.9 years (from 18 to 76 years). Of these, 12 were invasive pituitary adenomas and 42 were non-invasive pituitary adenomas. Immunohistochemistry showed that the expression of MMP 2 and MMP 9 in invasive pituitary adenomas was significantly higher than that in non-invasive pituitary adenomas (3.9 ± 0.5; 2.3 ± 0.2; P <0.01) and (4.1 ± 0.4; 2.6 ± 0.2; P <0.01). There was no significant correlation between the expression of MMP 2 and MMP 9 and the expression of MIB 1 (r = $ C 0 .0 5; P> 0.05). The expression of MMP 2 and MMP 9 mRNA in invasive pituitary adenomas was significantly higher than that in non-invasive pituitary adenomas [(5 9.7 ± 12.5)%; (33.3 ± 5.4)%, P <0.05) and [(6 8.2 ± 15.3)%; (2 1.8 ± 8.2)%; P <0.05]. Conclusion The high expression of MMP 2 and MMP 9 is closely related to tumor invasion. But no significant relationship with tumor size and secretory function.
