目的对比有饲养层的有血清培养体系(feederlayer,serum—containingmedium ,FSCM)和无血清培养体系(serumfreemedium ,SFM)培养人角朊细胞(keratinocyte ,KC)的特征。方法以两种不同培养体系培养人KC ,比较2 4h细胞贴壁数、完全汇合时间和最终分化形式。结果KC在两种不同培养体系中的形态、2 4h细胞贴壁数和完全汇合时间无统计学差异。FSCM体系能促使角朊细胞发生终末分化;SFM体系内角朊细胞增殖较FSCM体系快,但未发生终末分化。结论有饲养层的有血清培养体系培养人角朊细胞形成复层膜片,适合临床应用;无血清培养体系培养人角朊细胞形成单层细胞,适合KC生物学性征的实验研究。
Objective To compare the characteristics of cultured human keratinocytes (KCs) with feeder layer, serum-containing medium (FSCM) and serum free medium (SFM). Methods Human KCs were cultured in two different culture systems. The number of adherent cells, the confluence time and the final differentiation pattern were compared at 24 h. Results There was no significant difference in the morphology of KC in two different culture systems, the number of adherent cells at 24 h and the complete confluent time. FSCM system can promote terminal differentiation of keratinocytes; SFM system, keratinocyte proliferation faster than the FSCM system, but did not occur terminal differentiation. Conclusions There is a feeder layer serum culture system to culture human keratinocytes to form a multilayered membrane suitable for clinical application; serum-free culture system to culture human keratinocytes to form monolayer cells, suitable for KC biological characteristics of the experimental study.
