目的 检测小鼠ARL-1同源蛋白的表达情况,分析人醛糖还原酶相似基因(ARL-1)与小鼠ARL-1同源蛋白的同源性和遗传距离,从进化的角度讨论小鼠ARL-1同源蛋白与人ARL-1的亲缘关系。方法 利用OenBank和Swiss-Prot数据库,采用Clustal X 1.8软件分析人ARL-1与小鼠ARL-1同源蛋白的同源性,利用Mega 2.0软件分析人ARL-1与小鼠ARL-1同源蛋白的遗传距离,推测其进化关系;采用westernblot方法,运用抗ARL-1多克隆抗体分析小鼠多种组织ARL-1同源蛋白的表达情况。 结果 人ARL-1分别与小鼠中国仓鼠卵巢还原酶(CHO-Red)、成纤维细胞生长因子调节蛋白(FR-1)、鼠醛糖还原酶相似蛋白(rARLP)、小鼠输精管蛋白(MVDP)、鼠晶状体醛糖还原酶(LeAR)、δ4-3-5β酮甾类还原酶(5β-Red)、鼠醛酮还原酶蛋白C(RaK-c)、3α-18-羟甾类脱氢酶(3α-HSD)有83%、82%、81%、79%70%、51%、50%、45%的同源性。人ARL-1与小鼠CHO-Red的遗传距离最短(18.0%,P=0.023),其次是FR-1(19.1%,P=0.023)和rARLP(19.9%,P=0.025)。从进化树来看,人ARL-1与小鼠FR-1、rARLP、CHO-Red亲缘关系较近,其次是MVDP与LeAR。采用western blot在小鼠输精管,睾丸、膀胱、子宫检测到ARL-1同源蛋白。 结论 人ARL-1与小鼠CHO-Red、FR-1、rARLP、MVDP有较高的同源性、较?
Objective To detect the expression of ARL-1 homologous protein in mice and to analyze the homology and genetic distance between ARL-1 analogue gene of human aldose reductase and mouse ARL-1 homologue protein. Methods The homologies between human ARL-1 and mouse ARL-1 homologues were analyzed using OenBank and Swiss-Prot databases using Clustal X 1.8 software. The genetic distance between human ARL-1 and mouse CHO-Red was the shortest (18.0%, P = 0.023), followed by FR-1 (19.1%, P = 0.023) and rARLP (19.9%, P = 0.025). From the perspective of phylogenetic tree, human ARL-1 has close relationship with mouse FR-1, rARLP and CHO-Red, followed by MVDP and LeAR.
