目的:研究平糖煎对2型糖尿病大鼠血清及组织蛋白肿瘤坏死因子-α(TNF-α)、单核细胞趋化因子(MCP-1)含量的影响,探讨平糖煎治疗2型糖尿病的作用机制。方法:用高热量饲料加以链脲佐菌素腹腔注射的方法复制大鼠糖尿病模型,西药组给予二甲双胍,中药组给予平糖煎,用药4周后利用ELISA法与Western Blot法检测大鼠血清及组织蛋白中TNF-α、MCP-1含量变化情况。结果:利用ELISA法与Western Blot法检测TNF-α、MCP-1血清与组织含量变化情况,结果显示平糖煎组和西药组均好于模型组(P<0.01),而MCP-1利用ELISA法与Western Blot法检测结果显示平糖煎组与西药组比较,平糖煎组好于西药组(P<0.05)。结论:平糖煎对TNF-α、MCP-1等炎性因子能下调蛋白表达,有良好的调节作用,提示平糖煎和二甲双胍具有抗炎作用。
Objective: To study the effects of Pingtangjian on the levels of serum tumor necrosis factor-α (TNF-α) and monocyte chemoattractant protein (MCP-1) in type 2 diabetic rats and to explore the effects of Pingtangjian on type 2 diabetes mellitus The mechanism of action. Methods: The model of diabetic rats was established by intraperitoneal injection of streptozotocin (STZ) with high caloric diet. Metformin was given to the western medicine group and the plain sugar was given to the Chinese medicine group. After 4 weeks of treatment, the serum and Changes of TNF-α and MCP-1 in tissue proteins. Results: The changes of serum and tissue contents of TNF-α and MCP-1 by ELISA and Western Blot showed that the levels of TNF-α and MCP-1 in model group were significantly higher than those in model group (P <0.01) The results of the method and Western Blot test showed that the level of sugar in the group of Jiancang was better than that in the group of western medicine (P <0.05). CONCLUSION: Pingtangjian can inhibit the expression of TNF-α and MCP-1 and other inflammatory cytokines and down-regulate the protein expression, which implies that the anti-inflammatory effects of Pingtang Jian and metformin are good.
